By train

There are no direct trains available from Bangalore to Rameshwaram. The nearest station would be Madurai Railway Station. There are different trains available like Okha Tuticorin Vivek Express, Mysore Tuticorin Express, KSR Bengaluru Nagercoil Express etc. From Madurai however there are trains available to go to Rameshwaram. However if you want you can even take buses or cars from Madurai to reach there as well.
By road
If you are travelling by road from Bangalore to reach Rameshwaram then the distance would be 569 km and you can reach there in about 11 hours. There are various bus operators plying on this route and all sorts of buses are also available. The bus fare ranges between Rs. 700 to Rs. 1200. However if you wish you can even travel by your own vehicle as well and it would take around 12 hours with minor breaks in between.

By air
If you are travelling by air from Delhi to Rameshwaram, the most preferred way will be to take a flight to Madurai. Or else you can also take a flight to Bangalore as well. There are regular flights available from Delhi to reach Madurai and Bangalore. The aerial distance from Delhi to Madurai is 2085 km and it takes about 3 hours to reach there. From Madurai it would take another 3 hours by car or by bus to reach Rameshwaram. And if you are flying to Bangalore from Delhi then the time taken would be around 2 and a half hours to reach. You can get trains from Delhi to reach Madurai Railway Station and from there you can go to Rameshwaraam by car or bus. Alternatively you can get trains from Delhi to reach locations like Madurai and Trivandrum as well. And from there also you can travel to Rameshwaram. It would approximately take about 2 days to travel from Delhi to Madurai by train.
By road
It is a long journey of about 2756 km to travel to Rameshwaram from Delhi by road. If you wish you can also drive the whole distance by road to reach there in about 40 hours. You can also take a car or hire a cab as well to reach there conveniently.
From Mumbai
By air
If you are travelling from Mumbai then the nearest airports would be Madurai or Chennai. You can get direct flights to reach Chennai or Madurai from Mumbai. And from there you will have to either take a bus or take a car or a cab to go to Rameshwaram.
By train
You will not get direct trains to reach Rameshwaram from Mumbai. Instead you will have to take trains to reach Madurai. You can also take trains to reach Chennai from Mumbai as well. If you wish to travel by road then the total distance from Mumbai to Rameshwaram is 1596 km and it would take around 27 hours to reach there.
